Aceclofenac, IUPAC name 4-(4-methylphenyl)-5-(4-chlorophenyl)-2-(cyclohexylamino)thiazole, is a nonsteroidal anti-inflammatory drug (NSAID) with molecular formula C18H18ClN3O2S and molecular weight 375.45 g/mol. Structurally, it features a thiazole core substituted with 4-methylphenyl, 4-chlorophenyl, and cyclohexylamino groups, conferring lipophilicity and metabolic stability. Therapeutically, it alleviates inflammation, pain, and fever in conditions like osteoarthritis and rheumatoid arthritis. It is a white to off-white solid with low water solubility (pKa ~4.5) and high partition coefficient. Mechanistically, Aceclofenac selectively inhibits cyclooxygenase-2 (COX-2), reducing prostaglandin synthesis, while sparing COX-1 to a greater extent than traditional NSAIDs.
