Avanafil, IUPAC name 2-amino-6-(1H-tetrazol-5-yl)-4-[(5-chloro-2-oxo-1,3-benzothiazol-3(2H)-ylidene)methyl]pyrimidin-5-yl]thio]benzamide, is a pyrimidine-based phosphodiesterase type 5 (PDE5) inhibitor. Molecular formula C13H11ClN6OS (molecular weight 334.77 g/mol). It features a pyrimidine core with a tetrazole and benzothiazole moiety. Therapeutically used for erectile dysfunction. It is a white to off-white solid with low water solubility and high lipophilicity. Avanafil selectively inhibits PDE5, enhancing cGMP levels in penile smooth muscle, promoting vasodilation. Rapid onset of action due to high bioavailability and metabolic stability.
