Baclofen, chemically known as 4-Aminocrotonic acid (C4H8NO2, molecular weight 102.11 g/mol), is a gamma-aminobutyric acid (GABA) derivative characterized by a substituted butyric acid backbone with an amino group at the gamma position. It is a white, crystalline, water-soluble compound with a pKa of approximately 4.5. Clinically, it is employed to manage spasticity associated with multiple sclerosis, spinal cord injuries, and cerebral palsy. Mechanistically, Baclofen acts as a GABA-B receptor agonist, potentiating inhibitory neurotransmission in the spinal cord to reduce muscle hyperexcitability. Its lipophilic nature facilitates central nervous system penetration, while its short half-life necessitates frequent dosing.
