Canagliflozin, IUPAC name 1-[(3R,5S)-3,5-dihydroxy-4-(hydroxymethyl)oxolan-2-yl]-4-(propan-2-yl)pyridin-2(1H)-one, is a sodium-glucose cotransporter 2 (SGLT2) inhibitor with molecular formula C13H19NO6 and molecular weight 285.3 g/mol. It belongs to the pyridinone class, featuring a hydroxymethyl-substituted oxolane ring and a branched isopropyl group. Therapeutically, it is used for managing hyperglycemia in type 2 diabetes mellitus. It is a white to off-white solid, slightly soluble in water, with a pKa of ~4.5. Its mechanism involves competitively inhibiting SGLT2 in renal proximal tubules, reducing glucose reabsorption and promoting urinary excretion.
