Celecoxib, IUPAC name 4-[5-(4-methylphenyl)-3-(trifluoromethyl)-1H-pyrazol-1-yl]benzenesulfonamide (C17H14F3N3O2S, molecular weight 381.38 g/mol), is a sulfonamide-derived COX-2 inhibitor. Its structure features a pyrazole ring, trifluoromethyl group, and sulfonamide pharmacophore. Therapeutically, it targets inflammation and pain by selectively inhibiting cyclooxygenase-2 (COX-2), sparing COX-1. Physicochemically, it is a white solid with low water solubility (logP ~3.5) and moderate lipophilicity. Mechanistically, it binds reversibly to the COX-2 active site, blocking prostaglandin synthesis. Its selectivity arises from differential binding to COX-2 over COX-1 isoforms.
