Clofarabine, chemically 2-chloro-9-(2-deoxy-ฮฒ-D-ribofuranosyl)adenine, is a purine nucleoside analog with molecular formula C10H11ClN4O4 and molecular weight 286.45 g/mol. It features a 2-chloro-substituted adenine base linked to a 2-deoxyribose sugar. Therapeutically employed in treating pediatric acute lymphoblastic leukemia (ALL), it exhibits water solubility and a pKa of approximately 5.8. Mechanistically, it is phosphorylated intracellularly to its triphosphate metabolite, which competitively inhibits DNA synthesis by incorporating into DNA, inducing chain termination and apoptosis in proliferating cells.
