Dianhydrovalrubicin, IUPAC name 1,3-dianhydro-4,5,6,7,8,9-hexahydro-4,6-dihydroxy-5,8-dimethyl-2H-indeno[1,2-b]pyrrolo[3,4-e]pyridin-2-one, is an anthracycline derivative with molecular formula C27H25NO5 (molecular weight 443.44 g/mol). It features a tetracyclic anthracycline core with a dianhydro sugar moiety and a valeryl side chain. Therapeutically, it acts as a prodrug metabolized to valrubicin, an antineoplastic agent used in hematological malignancies. It is a yellow crystalline solid with low water solubility and high melting point. Mechanism of action involves DNA intercalation and topoisomerase II inhibition, inducing apoptosis via DNA damage.
