Dolutegravir, (5S,12aR)-5-(2,4-difluorophenyl)-4,5,6,7,8,9,10,11-octahydro-2H-indeno[5,4-b]pyridine-3-carboxamide (C18H18F2N4O; MW 344.36 g/mol), is an integrase strand transfer inhibitor (INSTI) with a difluorophenyl-substituted indenopyridine scaffold. It is therapeutically employed in antiretroviral therapy for HIV-1 treatment. The white to off-white solid exhibits high aqueous solubility and metabolic stability. Mechanistically, it binds to the viral integrase enzyme, blocking viral DNA integration into host chromosomal DNA, thereby inhibiting HIV replication.
