Fulvestrant, IUPAC name 3-(4-hydroxyphenyl)-N-(2-hydroxyethyl)-1-propyl-1H-indazole-5-carboxamide, is a selective estrogen receptor downregulator (SERD) with molecular formula C18H19NO3 and molecular weight 309.35 g/mol. It features an indazole core, aromatic hydroxyl groups, and an amide linkage. Therapeutically, it targets estrogen receptor-positive breast cancer by irreversibly degrading estrogen receptors, inhibiting tumor growth. It is a white to off-white solid with low water solubility and high lipophilicity. Mechanistically, it binds to estrogen receptors, inducing conformational changes that promote receptor ubiquitination and proteasomal degradation, thereby blocking estrogen signaling.
