Gestodene EP Impurity B is a structurally related synthetic byproduct arising from the partial hydrogenation of the parent compound's A-ring during Gestodene synthesis. It retains the 19-nor-17ฮฑ-pregnane scaffold but features a fully saturated cyclohexane ring in place of the aromatic A-ring, coupled with a 17ฮฒ-hydroxy group and a 3-oxo-ฮ4 moiety. This impurity lacks the conjugated double bond critical for Gestodene's progestogenic activity, rendering it inert pharmacologically. Its distinct A-ring saturation enables differentiation via UV/Vis and NMR spectroscopy. Analytically, it serves as an HPLC reference standard for quantifying process-related impurities in Gestodene drug substance batches.
